Fluorous Microwave Palladium CouplingFTI scientists have shown that the combination of a microwave accelerated reaction with a fluorous facilitated separation can dramatically increase the efficiency of high-speed organic synthesis. Palladium-catalyzed cross-coupling reaction of aryl perfluoroalkylsulfonates (C8F17O2SOAr) and thiols (RSH) under microwave irradiation afforded aryl sulfides which were purified by fluorous solid-phase extraction (F-SPE). No fluorous solvents are involved in the reaction and separation process. This method of fluorous synthesis has been further extended to multi-step syntheses of substituted hydantoin and amide scaffolds. FTI now offers the Pd(dppf)Cl2 reagent used below in a fluorous form. Cutting Edge Fluorous ChemistryFluorous CDMT and Solution-Phase Peptide Synthesis: Recently, Marcin Markowicz and Roman Dembinski of Oakland University reported the use of a fluorous CDMT coupling reagent in the synthesis and purification of peptides produced by solution-phase techniques.[1] CDMT has been reported as a condensing reagent superior to DCC and HOBT in syntheses involving alpha, alpha-disubstituted amino acids.
